Methylation analysis of Igf2 / H19 Imprinting Control Region (ICR) in Type II Diabetes Mellitus induced mice using Methylation-Specific PCR

چکیده
The imprinting control region (ICR) is a that controls imprinted expression of Igf2/H19 so Igf2 will only be expressed from the paternal allele instead maternal allele. This research aimed to reveal if ICR methylation status' change contributes emergence type II diabetes Mellitus (T2DM) throughout induction in mice model. changes were detected by methylation-specific PCR (MSP). Control group was given normal diet and buffer vehicle injection while treatment high fat (HFD) six times 60 mg/kg body weight streptozotocin injection. process conducted for 12 weeks. insulin tolerance test (ITT) on week 0, 9, 12. treatment's successfully induced significantly higher area under curve (AUC) compared (P < 05). Moreover, ITT showed diabetic had glucose levels at every time point test. Meanwhile, there no evident through 0 Thus, not proven contribute T2DM.

The Igf2/H19 imprinting control region exhibits sequence-specific and cell-type-dependent DNA methylation-mediated repression
Methylation of CpGs is generally thought to repress transcription without significant influence from the sequence surrounding the methylated dinucleotides. Using the mouse Igf2/H19 imprinting control region (ICR), Igf2r differentially methylated region 2 (DMR2) and bacterial sequences, we addressed how methylation-dependent repression (MDR) from a distance varies with CpG number, density and su...

Background Genomic imprinting is a heritable and developmentally essential phenomenon by which gene expression occurs in an allele-specific manner1. While the imprinted alleles are primarily silenced by DNA methylation, it remains largely unknown how methylation is targeted to imprinting control region (ICR), also called differentially methylated region (DMR), and maintained. Here we show that ...

Backgrund: Imprinted genes are a unique subset of few genes, which have been differentially methylated region (DMR) in a parental origin-dependent manner during gametogenesis, and these genes are highly protected during pre-implantation epigenetic reprogramming. Several studies heve shown that the particular vulnerability of imprinting genes during suboptimal pre- and peri-conception microenvir...

Background: To investigate if aberrant methylation and expression of imprinted genes of the Igf2-H19 locus in the spermatozoa and embryos could be a paternal epigenetic factor involved in early embryo loss To elucidate the role of estrogen in acquisition of the imprinting at the Igf2-H19 locus during spermatogenesis Materials and Methods: Adult male rats of Holtzman strain were administered tam...
متن کاملCpG methylation regulates the Igf2/H19 insulator
The differentially methylated 5'-flank of the mouse H19 gene unidirectionally regulates the communication between enhancer elements and gene promoters and presumably represses maternal Igf2 expression in vivo [1-6]. The specific activation of the paternally inherited Igf2 allele has been proposed to involve methylation-mediated inactivation of the H19 insulator function during male germline dev...
